Calcium controls Sleep duration in Mice

Summary:Researchers have identified seven genes responsible for causing mice to stay awake or fall asleep based on a theoretical model of sleep and on experiments using 21 different genetically modified mice, some of which showed different sleep durations. Researchers hope that their research will contribute to the understanding and treatment of sleep disorders and associated neurodegenerative diseases.

University of Tokyo and RIKEN researchers have identified seven genes responsible for causing mice to stay awake or fall asleep based on a theoretical model of sleep and on experiments using 21 different genetically-modified mice, some of which showed different sleep durations. Researchers hope that their research will contribute to the understanding and treatment of sleep disorders and associated neurodegenerative diseases.

All animals appear to sleep for varying durations. In humans, sleep duration changes over our lifetime, gradually reducing from birth through to old age. The mammalian body appears to use time spent asleep to recover from the effects of daily life, such as removing waste products from the brain and restoring the immune system, and may use the time to process experiences and lay down long-term memories. However, the fundamental reasons for sleep and the mechanisms by which sleep duration is regulated remain largely unknown.

Professor Hiroki Ueda and his research group at the University of Tokyo’s Graduate School of Medicine and the RIKEN Quantitative Biology Center developed a computational model of sleep, based on which they predicted that sleep duration is regulated by calcium and identified multiple genes that were potentially involved. The research group then tested their predictions against 21 different genetically modified mouse types to reveal that a mechanism regulated by calcium ions is indeed responsible for controlling sleep duration.

The research group applied an efficient type of the CRISPR method developed in earlier research to remove genes involved in calcium regulation to create 21 genetically modified mice. Out of the 21, seven exhibited significant changes in sleep duration. In addition, the research group also showed that the inflow of calcium ions into neurons is required for mice to fall asleep and that pumping calcium ions out of neurons is required for mice to wake up. These results were in line with their predictions from the computational model.

“We were thrilled that our experimental data backed up the predictions of our model so well,” says Fumiya Tatsuki, the first author of the paper. He continues, “Our recently developed “snappy sleep stager,” which monitors breathing to measure sleep duration without harming the mice, was an essential tool for evaluating whether our genetically modified mice exhibited changes in sleep duration.”

“Sleep is one of the most fundamental physiological functions. From flies to humans, it seems that most animals sleep, but we still know so little about the molecular processes by which sleep duration is regulated. This research has provided us with a wide range of potential targets for future research and therapies,” says Ueda. “In addition, many mental disorders and neurodegenerative diseases are also associated with sleep disorders, so understanding how sleep is regulated may lead to a deeper understanding of those diseases and future treatments.”

The Water Storage Tank with a capacity of 150,000 litres will benefit 10,000 villagers of Padghe Village, which is situated near the Company’s High Throughput Barrel Manufacturing Plant at Taloja, Navi Mumbai. This facility will play an important role in reducing the problem of water supply in the village in turn improving sanitation and quality of life of the people. The implementation partner, Rotary Club of Industrial Town, Panvel, executed the project on time.

Balmer Lawrie’s CSR initiatives are founded on two Flagship Schemes Balmer Lawrie Initiative for Self Sustenance [BLISS] and Samaj Mein Balmer Lawrie [SAMBAL]. The first scheme is directed at providing and improving the long term economic sustenance of the underprivileged. New initiatives to combat TB launched

The Government of India stands committed to accelerating its efforts to combat tuberculosis in the country”, stated Shri J P Nadda, Union Minister of Health and Family Welfare. Speaking on the eve of ‘World TB Day’, he stated that the process of fighting TB is continuous, hence there can be no dilution and do diversion. Our attention needs to steadfast and aggressive, stated the Health Minister. He further added that resources will not be a constraint and the Government will continue to work with all stakeholders, in devising short term and long term approaches. Shri Nadda also emphasized the need for compassion in the treatment of TB patients.

At the event, Shri J P Nadda, launched Bedaquiline – new anti-TB drug for Drug Resistant TB as part of the RNTCP. The drug is a new anti-TB drug for treatment of MDR-TB. This new class of drug is a diarylquinoline that specifically targets Mycobacterial ATP synthase, an enzyme essential for supply of energy to Mycobacterium tuberculosis and most other mycobacteria. This drug is indicated for use in the treatment of drug-resistant TB. Bedaquiline is being introduced at six identified tertiary care centres across India. These sites have advanced facilities for laboratory testing and intensive care for patients. Bedaquiline will be given to multi-drug resistant TB patients with resistance to either all fluoroquinolone and/or all second line injectables and extensive drug resistant TB.

Shri Nadda also inducted over 500 Cartridge Based Nucleic Acid Amplification Test (CBNAAT) machines in the programme. The CBNAAT is a revolutionary rapid molecular test which detects Mycobacterium tuberculosis and rifampicin drug resistance, simultaneously. This test is fully automated and provides results within two hours. It is a highly sensitive diagnostic tool and can be used in remote and rural areas without sophisticated infrastructure or specialized training. Until 2015, 121 CBNAAT sites are functional in the country largely providing decentralized testing for detection of DR TB. With the availability of these additional 500 machines, access to rapid quality assured diagnosis of DR TB and TB will be ensured in all the districts of India either directly or through a linkage by specimen transport mechanism. Additionally, the programme will be able to use this highly sensitive state-of-art technology for diagnosis of TB among key populations like children, PLHIV and extra pulmonary TB patients.

The Health Minister also released the TB India 2016 Annual Report and the Technical and Operational Guidelines for TB Control in India 2016. The comprehensive set of guidelines are for management of all forms of TB, including drug resistant TB and strategies for intensified case finding; adherence of more patient centric approaches; and single-window care for patients suffering from HIV and TB. A handbook for ‘Healthcare Worker surveillance for TB in India’ and Guidelines for Prevention and Management of Adverse reactions associated with anti-TB drugs were the other e-book launch at the event. A new radio campaign with TB ambassador Amitabh Bachchan and social media campaign, was also launched at the event.

The Minister also launched the ‘Third line ART programme for People Living with HIV’. The life-saving third line ART costs nearly Rs. 1.18 lakh per patient per year. Providing these free would not only safe lives but improve socioeconomic conditions of the patients. This initiative brings India’s ART programme at par with programmes in the developed countries.

Shri B P Sharma, Secretary (HFW) highlighted the need for collective commitment of all stakeholders. He stated that the Revised National Tuberculosis Control Program (RNTCP) is one of the most successful programmes. “The programme has made significant impact on prevalence and treatment of TB”, said Shri B P Sharma. He further added that 300 CBNAAT machines have already been installed and 200 more will be installed soon. “Quality of treatment has to be even over the public and private sectors. It has to be well supported by a strong procurement so that it can be sustained till 2030, said Shri B P Sharma. Secretary (Health) also laid emphasis on surveillance and monitoring. He said that we need new tools for diagnostics and new research. He further said that delivery mechanism should be in conformity with goals we have set for ourselves.

Speaking on the occasion, Dr Saumya Swaminathan, Secretary (DHR) and DG (ICMR), highlighted the challenges of the frontline health workers. She said that there is a need for new and innovative tools for conducting tests and investment in research. She further added that we need to use drugs optimally.

Also present on this occasion were, Dr. (Prof) Jagdish Prasad, DGHS, Shri C K Mishra, AS & MD, and other senior officers of the Health Ministry, representatives of WHO, World Bank and other development partners.

Common painkillers more dangerous than We think

Summary:New research shows that common painkillers (NSAIDs) have more side effects than we think, and that they should be used with great care in patients with heart disease. NSAIDs are among the most sold medicines in the world.

Many patients are prescribed NSAIDs for the treatment of painful conditions, fever and inflammation. But the treatment also comes with side effects, including the risk of ulcers and increased blood pressure. A major new study now gathers all research in the area. This shows that arthritis medicine is particularly dangerous for heart patients, and also that older types of arthritis medicine, which have not previously been in focus, also appear to be dangerous for the heart.

“It’s been well-known for a number of years that newer types of NSAIDs — what are known as COX-2 inhibitors, increase the risk of heart attacks. For this reason, a number of these newer types of NSAIDs have been taken off the market again. We can now see that some of the older NSAID types, particularly Diclofenac, are also associated with an increased risk of heart attack and apparently to the same extent as several of the types that were taken off the market,” says Morten Schmidt, MD and PhD from Aarhus University, who is in charge of the research project. He adds:

“This is worrying, because these older types of medicine are frequently used throughout the western world and in many countries available without prescription.”

Each year, more than 15 per cent of the population in western countries are given a prescription for NSAIDs. This figure increases with age. Sixty per cent of the adult population in Denmark collects at least one prescription for an NSAID within a ten-year period. Heart patients are no exception and previous studies have shown that up to forty per cent of Danish patients with heart failure or previous heart attacks are prescribed NSAIDs.

The study, which was carried out in collaboration between 14 European universities and hospitals, including a number of leading European heart specialists, is today being published in the most prestigious European journal of heart medicine, European Heart Journal.

New guidelines

In the study, the researchers have gathered all research on the use of NSAIDs in patients with heart disease. The survey means that the European Society of Cardiology has now for the first time formulated a number of recommendations about what doctors should consider before prescribing painkillers to their patients.

“When doctors issue prescriptions for NSAIDs, they must in each individual case carry out a thorough assessment of the risk of heart complications and bleeding. NSAIDs should only be sold over the counter when it comes with an adequate warning about the associated cardiovascular risks. In general, NSAIDs are not be used in patients who have or are at high-risk of cardiovascular diseases,” says another of the authors, Professor in cardiology Christian Torp-Pedersen, Aalborg University, Denmark.

Consumption should be reduced even more

For a number of years, Danish researchers have made significant contributions in the field. One result of this work has been a reduction in the use of Diclofenac in Denmark. However, according to Morten Schmidt, there is still room for improvement:

“Many European countries consume more of these drugs than Denmark. But we can still do better and it’s often the case that paracetamol, physiotherapy, mild opioids or other types of NSAIDs with less risk for the heart would be better for the patients. Of course, the recommendations that have been introduced following our study and its review of the heart-related risks are a big step in the right direction in relation to patient safety,” says Morten Schmidt.

FACTS about arthritis medicine (NSAID):

  • NSAIDs is an abbreviation for Nonsteroidal Anti-Inflammatory Drugs and is used to treat a wide range of diseases, in particular disorders in the muscular and bone system, where the drug counteracts swelling, pain and limitations in movement associated with inflammation.
  • NSAIDs are not antibiotics and therefore do not help to fight infections caused by bacteria.
  • NSAIDs are in Denmark sold both in low doses (Ibuprofen 200 mg/tablet) without a prescription and in higher doses and other types with a prescription.

Shri Goyal was accompanied by Shri Keshaw Prasad Maurya MP (Phoolpur),  The dignitaries were welcomed by Shri S C Pandey, Director(Projects), NTPC, Shri Prabhat Kumar, CEO, Meja Urja Nigams and senior MUNPL officials.

During his visit, Shri Goyal inspected the power plant and reviewed its activities and performance. During his visit he also inaugurated a Community centre and told that more than 4000 people will be directly benefitted by the community centre . Addressing the media persons Shri Goyal stated that the Meja Project will also work to utilise sewage water for plant activities, he also assured  that Meja team for coal linkage from nearest coal mines,so the rate of electricity gradually decrease and it will benefit the people of Uttar Pradesh by reducing the per unit electricity charge project work for linkage of coal through nearest coal mines. He also said the Heart of the Power Plant is very impressive and he was particularly happy at the enthusiasm of the Team and the innovative work being developed for use in other Stations also. The dignitaries also planted saplings near the power plant.

It will facilitate efficient and cost-effective services like Cargo aggregation & dis-aggregation, Inter-Modal-Transfer, Sorting, Packing & Re-packing and Value-added-services, that will make InterModal-Transfer to Rail attractive in the supply chain of various types of customers. Besides having infrastructure to cater to the up-coming Dedicated Freight Corridor (DFC) in future, this MMLP will also be looking at handling wide variety of services such as Tank Farms for Liquid Cargo, Auto-Car Rakes, Specialized Warehouses for Value-Added services, Silos and customized facilities for cargo such as Cement, Food Grains etc.

‘Make in India’ companies to dominate at Defence Expo 2016, with 498 Indian companies confirming their participation to the Defence Exhibition Organisation (DEO).

Of the nearly 977 companies turning out from 46 nations (including India) at Goa this year, nearly 51 per cent are domestic exhibitors. The figures of the domestic participants have near doubled since the 2014 edition, in which there were 256 Indian exhibitors and 368 foreign stalls.

Among the familiar list of Indian exhibitors include Ashok Leyland, Bharat Electronics Limited, Bharat Forge, BrahMos Aerospace, Defence Research Developmental Organization, Goa Shipyard, Mahindra group and the Kirloskar’s, among the familiar names in the domestic defence circuit.

The Indian delegation of exhibitors in the pavilion is followed 479 foreign participants of whom 91 companies will represent from the United States, 71 from Russia, 46 companies from United Kingdom and 38 business ventures each from France, Germany and Israel. In comparison to 30 foreign nations participating in 2014, this year 46 nations will take part. However, China and Pakistan will not partake in the exhibition. “Although, the Chinese delegation maybe present,” Defence Minister Manohar Parrikar had said on earlier occasion.

Revival of 9 stranded Gas based Power Generation Plants

The Government is delighted to announce the revival of 9 stranded gas based power generation plants with an installed capacity of 5942 MW which have successfully bid through a transparent and competitive reverse e-auction process. These plants would generate 6.79 billion units of electricity which will be supplied at or below Rs 4.70 per unit to the purchaser Discoms during the period from1st April 2016 to 30th September 2016. The auction process successfully concluded on March 20, 2016 wherein 9 plants emerged as preferred bidders and were allocated 7.62 MMSCMD e-bid RLNG.

Previously an e-auction was wherein bidders participated in reverse e bidding process with Per unit PSDF ceiling price of Rs 0.41/unit. However, the Bidders did not get an opportunity to bid below Rs. 0.00 (Rupees zero only) per Unit, as option for the same was not built in. In light of the fact that the Bidders were not provided such an opportunity and the sufficient competition might not have been generated, a re–auction was conducted.

The re-auction resulted in aggressive negative bidding with the lowest bid at RS 0.03 per unit. As a result there is estimated savings of Rs 18.29 crore to Government’s Power System Development Fund.

The present reverse e-auction for the Stranded Gas based Plants is the third phase of auctions conducted under Scheme for Utilization of Stranded Gas Based Generation Capacity.

The grid connected gas based power generation capacity in the country is 24,150 MW. Of this, a capacity of 14,305 MW had no supply of domestic gas. These comprise 29 plants which were eligible to participate in the auction process. 11 plants with a cumulative installed capacity of 6626 MW participated in the bidding for Phase III.

The successful bidders include power plants in Southern region. The generation from these plants would improve the power availability in the Southern grid, mitigating to a large extent the shortage of availability and constraint of evacuating the power to the Southern region from other regions.

Government of India is committed to the revival of such stranded power plants as a measure to ensure 24X7 power to all.

Ms Namita Suhag, President Army Wives Welfare Association (AWWA) released a special edition of AWWA Journal, “Manasvini” during a workshop on the theme ‘Women Health and Lifestyle’ conducted at NCC Auditorium in Delhi Cantonment to commemorate the Golden Jubilee of Army Wives Welfare Association (AWWA). The book includes detailed and extensive information for modern-day women on diverse topics like parenting, lifestyle, common women related diseases and cures, etc, aiming to serve as their lifelong companion. A total of approximately 1000 ladies attended the programme.

An extensive Medical Check-up Camp by Army Doctors was organised on the occasion for the wives of all serving Army personnel of Delhi. A number of other cultural events were conducted including programmes on ‘Beti Bachao’ highlighting the menace of female infanticide. Lectures and talks by prominent speakers and specialists with the aim of sensitising the women about the modern-day lifestyle, adapting lifestyle for healthy living, gender issues, parenting in modern day environment, pregnancy related issues, home remedies for ailments including use of ayurveda and beauty related issues, were also conducted.

In order to increase awareness about prevalent problems of Diabetes, Anaemia, Osteoporosis, Cancer, etc in women, lectures by specialist doctors were also organised to educate the women about symptoms of these diseases for early diagnosis and cure.

People cooperation required to Improve Quality of Forests

The Government has sought peoples’ cooperation to bring about an improvement in the quality of forests and to enhance the scientific management of forests. Emphasising the need to appreciate the true value of forest in the growth and development of our country, Minister of State (Independent Charge) of Environment, Forest and Climate Change, Shri Prakash Javadekar, said here that every person must plant at least 10-15 trees during one’s lifetime and care for the trees, so that trees can create an ‘Oxygen Bank’. He added that just as money is deposited in banks, each person must plant one tree after every five years to create the fixed deposit of an ‘Oxygen Bank’. He pointed out that one change that has been brought about in plantation of trees in nurseries is that plants that attain a height of upto 6 feet are planted to ensure a high survival rate. The Minister said that students are the agents of change, as it is the students who can nurture and enhance the sensitivity towards environment.

Speaking on the occasion, Minister of Environment and Forest of Delhi government, Shri Imran Hussain said that the Delhi government will spare no effort to make Delhi more green.

Shri Javadekar and Shri Hussain also planted trees on the occasion and gave away prizes to the children. Shri Javadekar also released a Heritage Map on the occasion. School children from schools around Asola Bhatti sanctuary, presented a street play (Nukkad Natak) highlighting the exploitation of Earth. The school children had also put up an exhibition to mark the occasion.

This year’s theme of International Day of Forest is ‘Forest and Water’ to raise awareness about how forests are the key of the planet’s supply of fresh water is essential for life.
